The enzymatic degradation of cynaroside (luteolin 7-O-glucoside) has been investigated in fresh and dried artichoke (Cynara scolymus L.) leaves. Plant and drug samples obtained from various sources differed considerably with regard to their ability to hydrolyse cynaroside during fermentation, and exogenous cynaroside when added to cell-free protein extracts. Extractable cynaroside 7-O-glucosidase was particularly high in artichoke leaves dried at 35° C. A β-glucosidase hydrolysing cynaroside was partially purified from the herbal drug Cynarae folium in 0.2 % yield by the combination of ammonium sulphate fractionation, hydrophobic interaction chromatography on Phenylsepharose 6FF, anion exchange chromatography on Source 30Q, gel filtration on Superdex 200 prep grade HR 16/60 and anion exchange chromatography on Resource Q. The molecular mass of the enzyme was about 160 kDa as determined by SDS PAGE. The role played by endogenous plant enzymes in stabilizing flavonoid glycosides during processing is discussed.
